Protein domains page
In the framework of the PROSITE project,
we are constructing generalized profiles in order to describe local
homology regions in proteins and make them searchable.
In the course of this work, we frequently notice previously undescribed
homology regions that occur in several different otherwise unrelated
proteins. These homology regions, which apparently can live in different
sequence contexts, frequently confer a specific functionality to the
proteins harbouring them.
In some cases, the function of regional homology domains can be inferred
by comparing the properties of the host proteins. Detection of these
domains in uncharacterized sequences might give valuable clues in assignment
of protein function. In other cases, where the biological role of a
local homology domain is not known, the delineation and comparison of
these regions might still be useful e.g. for finding conserved residues
as targets for mutagenesis or for structural studies.
Most of the domains we detect will be included into PROSITE as profile
entries. We will also pick a selection of these domains, either new
or significantly extended ones, for a more detailed description on

- The FHA domain [25-Apr-1996]
- A putative nuclear signalling domain found in protein kinases and
transcription factors. Keywords: fork head, DUN1, RAD53, SPK1,
cds1, MEK1, KAPP, NIPP1, Ki-67, fraH, KIAA0170.
- The death domain [25-Apr-1996]
- A heterodimerization domain present in several proteins involved in
apoptotic signal transduction and the NFkB pathway.

- The PLD domain [25-Apr-1996]
- A duplicated sequence motif occuring in phospholipases D and related
phosphatidyltransferases, as well as in several other proteins using a
ping-pong phosphotransferase mechanism.

- The G-protein
desensitation domain [25-Apr-1996]
- A common motif in terminating G-protein mediated signals.

- The UBA domain [25-Apr-1996]
- A sequence motif found in multiple enzyme classes of the

SNF1-related kinases, par-1, BS4.
- The UBX domain
- A sequence motif of unknown function, associated with UBA domains
and ub-related proteins. Keywords: UBA, FAF1, undulin, ubiquitin.
- The PCI domain
- A sequence motif occuring in several subunits of the 26S
proteasome, the translation initiation factor eIF3, the COP9 complex and other
multi-protein complexes. Keywords: Proteasome, COP9, eIF3, Ikappa-B

- The MPN domain
- A sequence motif occuring in several subunits of the 26S
proteasome, the translation initiation factor eIF3, and other multi-protein

- The rhodanese homology domain
- A conserved domain present in rhodanese (N- and C-terminal), in
the catalytic domain of Cdc25 phosphatase, in the non-catalytic domain of
various MAP-kinase phosphatase families, in the non-catalytic domain of
several ubquitin hydrolases and in a variety of pro- and eukaryotic stress
